Abstract
Anthropogenic impacts within the watershed threaten the survival of sheat catfish. Therefore, species of the genera Ompok and Kryptopterus should be conserved and domesticated to ensure their long-term persistence. Moreover, the accurate documentation of the existence and taxonomic validity of Ompok and Kryptopterus species is crucial. Accordingly, this study aims to assess the species diversity of the genera Ompok and Kryptopterus in the Mahakam River, East Kalimantan, Indonesia. The study was conducted between June and December 2014, with an update in August 2024. Fish samples were collected from the upstream and middle reaches of the Mahakam River. A total of 655 fish were biometrically examined. Scatter plots of the principal components are used to distinguish among types and identify the primary characteristics. In addition, COI gene sequence alignment, genetic variation analysis (SNP), genetic distance, and phylogenetic tree reconstruction via the Neighbor-Joining method were performed. As reference species, GenBank contains 19 partial COI gene sequences from the genera Ompok and Kryptopterus, along with the Notopterus notopterus gene sequence as an outgroup. The COI gene sequence of Silurus glanis (Siluridae) was also used as a reference. This study demonstrated that biometrics, morphology, and genetic analysis produce mutually supportive outcomes. A strong key for practical identification will be particularly valuable for clarifying distinctions among species, as well as for their conservation and sustainable use.
